Yup, you can wear an item just by double-clicking it from your inventory. If you can't wear something, check that your character meets the level and statistic requirements.

Try it! Press E, then take off an item by double-clicking it. Then, double-click the item from the item inventory (in case you forgot, press I) to put it back on.
